Does this mean that the change I clicked to apply does not get activated at all?

It means it won't get activated until you restart, so it will eventually be activated.

It actually depends on what you change to know if a restart is needed.

Are there some general rules Tarun?

I think that generally if it's done through the GUI, unless it says to otherwise restart; you don't need to.

But if you mess with anything registry related, you need to reboot so it can load it on next start.
